Phenotype Homozygous State Decreased expression of CD45 on CD19⁺ B cells loc/loc mice have normal numbers of B cells Cell intrinsic defect Slightly reduced levels of IgM⁺IgD⁻, IgM⁺IgD⁺ and IgD⁺CD21⁺ cells Reduced levels of B220loCD19⁺ cells but normal levels of B220⁺CD19⁺ in bone marrow 3 fold decrease in DP thymocytes 10 fold decrease in CD4 SP thymocytes 5 fold decrease in CD8 SP thymocytes DP thymocytes had 2.5 fold increase in expression of CD4 and CD8, cell intrinsic Co-receptor expression on SP T cells was increased 1.5 fold Dramatic decrease in CD8⁺CD44lo/med cells TCRβ is up-regulated on DP thymocytes and down regulated on mature T cells.
