Protein expression of altered gene: Nil
Genetic alteration of gene: A 2.0kb region, containing portions of exons 2 and 3, was deleted by the insertion of a neomycin selection cassette adjoined to the 5' end of an upstream mouse sequence (UMS) element. Northern blot analyses, using various probes, showed that transcription was not terminated by the UMS element. RT-PCR and sequence analysis showed that exons 2 and 3 and the intervening neo cassette had been spliced out in the majority of transcripts. The exon-skipped transcript was shown to be capable of producing protein in a cell-free system, however, protein was undetected via Western blot analysis of lysates obtained from homozygous mutant mice. Additionally, assays of MEF cell extracts showed an absence of autophosphorylation activity in homozygous mutant mice.

Phenotype Homozygous State Mice homozygous for disruptions in this gene display altered susceptibility to viral infection. Enlarged lymph nodes. Altered susceptibility to viral infection. increased T cell proliferation. Proliferative responses of both CD4+ and CD8+ T-cells from sensitized mice is somewhat increased abnormal inflammatory mediator physiology. A 2 fold increase in Il4 production by CD44+ cells increased susceptibility to viral infection. Susceptibility to encephalomyocarditis virus tested response to infection and survival were similar to control mice; however, pretreatment of mice with IFNgamma failed to improve survival as it did in controls increased susceptibility to type IV hypersensitivity reaction. Exaggerated response to challenge after presensitization in contact hypersensitivity test effect can be transferred to naive mice using CD8+ cells from sensitized mice.
