Strain nomenclature: C57BL/6JSfdAnu-Dock8cpm/AnuApb
Nickname: Captain Morgan, ENU4AT:038
Strain designation name: C57BL/6JSfdAnu-Dock8cpm/AnuApb
Genetics
Strain Types Mutant strain
Genetic Details Chemically induced : ENU
Mode of Inheritance Recessive
1.  Affected Gene Name: dedicator of cytokinesis 8
Chromosome 19
Affected Gene Symbol Dock8
Protein expression of altered gene: Nil
Genetic alteration of gene: ENU-induced mutation. The mutation comprises a T to C transition that modifies the splice donor sequence of exon 20, resulting in aberrant mRNA splicing between cryptic splice donors and the normal exon 21 splice acceptor. cDNA sequence analysis has identified the abnormal splice products: frameshifted mRNAs containing premature translation stop codons that terminate the protein before the DOCK-homology region 2 (DHR2) domain. DHR2 is known to bind to G proteins of the Rho family, promoting GTP -- GDP exchange and initiating a variety of cellular responses.
Location on chromosome (bp): 25074019-25276922 bp, + strand (From NCBI annotation of NCBI Build 37)

Version: NCBI m37, Ensembl build 57
Mutant Construction Technique None
Phenotype
Phenotype Homozygous State Defective antibody response to immunization: failure to sustain antibody formation, undergo affinity maturation or to mount heightened recall (memory) response upon booster immunization. DOCK8-mutant B cells were unable to form marginal zone B cells or to persist in germinal centers and undergo affinity maturation. Dock8 mutations disrupted accumulation of the integrin ligand ICAM-1 in the B cell immunological synapse but did not alter other aspects of B cell antigen receptor signaling.
Original Genetic Background C57BL/6JSfdAnu
Genetic Background Currently Maintained C57BL/6JSfdAnu
Strain Images
Captain morgan homozygous mutant mice (cpm/cpm), primurus homozygous mice (pri/pri), compound heterozygotes between the two strains (cpm/pri), and wild-type control mice (+/+) were immunized with chicken gammaglobulin (CGG) coupled to the hapten azobenzene arsonate (ARS). Antibodies to CGG and to ARS were measured in serum of individual mice (dots) by ELISA, 14 and 28 days after primary immunization. Antibody titres are shown relative to a reference immune serum pooled from wild-type mice.
