Genetic alteration of gene: ENU mutagenesis induced a point mutation (T to A chr 11: 96973058) in the second intron altering the splice acceptor site and thereby preventing normal splicing. This mutation deletes 8 amino acids from the final peptide. Activated splenocytes exhibit a 3- to 4-fold decrease in protein expression compared with similarly treated wild-type cells.

Phenotype
Phenotype Homozygous State Defective antibody response to immunization: failure to mount a normal Th1 polarized IgG2c antibody response to heat killed B pertussis bacteria. Few circulating NK cells. Decreased NK cell number: the percent of NK cells in the blood, spleen, and liver is lower than in wild-type mice. Increased NK cell number: * the frequency of NK cells in the peripheral lymph nodes and bone marrow is increased compared to in wild-type mice. * NK cells accumulate in lymph nodes due to defective egression. Absent NK T cells: mice exhibit a near complete absence of invariant natural killer T cells unlike in wild-type mice. Abnormal NK cell differentiation: * NK cells exhibit an immature phenotype unlike in wild-type mice. * NK cells accumulate in lymph nodes due to defective egression. * in reconstitution experiments, derived NK cells accumulate in the peripheral lymph nodes due to defective egression.

Animals from pedigree or wild-type C57BL/6 control stocks were immunized with chicken gammaglobulin (CGG) coupled to the hapten azobenzene arsonate (ARS), and heat-killed Bordetella pertussis bacteria, precipitated together in alum. IgG1 antibody to CGG and IgG2c antibody to B pertussis were measured in serum of individual mice (dots) by ELISA 14 days after primary immunization, showing normal primary Th2-polarized antibody formation to CGG but deficiency in mounting a Th1-polarized antibody response to B pertussis. Note that the animals fall in three phenotypic groups, corresponding to homozygous mutant, heterozygotes, and homozygous wildtype.

Identified in flow cytometry screen for deficiency of circulating NK cells by Dr Craig Jenne, Dr Susan Watson, Prof Lewis Lanier and Prof Jason Cyster (UCSF). Mutation mapped and identified by Dr Anselm Enders (JCSMR, ANU). Project guided by Prof Chris Goodnow.
