Phenotype
Phenotype Homozygous State Defective antibody response to immunization: failure to sustain antibody formation, undergo affinity maturation or to mount heightened recall (memory) response upon booster immunization. DOCK8-mutant B cells were unable to form marginal zone B cells or to persist in germinal centers and undergo affinity maturation. Dock8 mutations disrupted accumulation of the integrin ligand ICAM-1 in the B cell immunological synapse but did not alter other aspects of B cell antigen receptor signaling.

primurus homozygous mice (pri/pri), and heterozygous (pri/+) or wild-type (+/+) siblings were immunized with chicken gammaglobulin (CGG) coupled to the hapten azobenzene arsonate (ARS), and heat-killed Bordetella pertussis bacteria, precipitated together in alum. IgG1 antibody to CGG and IgG2c antibody to B pertussis were measured in serum of individual mice (dots) by ELISA 14 days after primary immunization, showing normal primary T-dependent antibody formation and polarization. After 37 days, the mice received a booster immunization of CGG-ARS and NP hapten coupled to ficoll, and 7 days later (d44) serum antibodies to CGG, ARS and NP were measured, showing failure to sustain CGG antibody, failure of affinity maturation to produce ARS antibody, but normal T cell independent type 2 antibody response to NP-ficoll.

Antibodies to CGG and to ARS were measured in serum of individual mice (dots) by ELISA, 14 and 28 days after primary immunization. Antibody titres are shown relative to a reference immune serum pooled from wild-type mice.
