Phenotype Homozygous State Constitutive expression of the IL5 gene is sufficient to induce lifelong high-level eosinophilia. Northern blot analysis identified 11,5 mRNA in the thymus, Peyer's patches, and subcutaneous (pooled inguinal and axillary) lymph nodes. No IL5 mRNA was detected in liver, heart, brain, or skeletal muscle tissues from transgenics, nor in any tissues from normal littermates. Do not maintain as homozygotes. While this is apparently possible, survival of homozygotes likely to be lower.
Phenotype Heterozygous State Appear normal with peripheral blood eosinophilia and increased numbers of eosinophils in the spleen (with splenomegaly), bone marrow. Heterozygous BALB/c and C57BL/6 IL-5 Tg2 mice are resistant to methylcholanthrene-induced carcinogenesis - see Simson, L, et al., 2007, Regulation of carcinogenesis by IL-5 and CCL11: a potential role for eosinophils in tumor immune surveillance. J. Immunol., 178:4222-4229.

How is this strain characterised? Phenotyping
Characteristics Assayed For We routinely phenotype IL-5 Tg mice by counting cells from tail blood smears, with eosinophils being the predominant cell type present if the mouse is IL-5 Tg. All mice that will be sent for preservation have been identified as IL-5 Tg through a blood smear taken at weaning and then again a few weeks later to confirm the IL-5 Tg status. We do not usually use PCR.

Additional Information IL-5 Tg mice have enhanced resistance to some parasitic helminths (Nippostrongylus brasiliensis, Strongyloides ratti), but not to others (Schistosoma mansoni, Trichinella spiralis, Toxocara canis). Expression of the IL-5 Tg on CBA/Ca, C57BL/6 and BALB/c genetic backgrounds enhances the immunopathology of aeroallergen-induced lung inflammation, but the CBA/Ca mice are relatively resistant to development of several aspects of the disease. Mammary tissue development is temporarily retarded in IL-5 Tg mice.
