Strain name: B6.Cg-Gzmatm1Simn Gzmbtm1Ley/Apb
Nickname: B6.gAB-/-
Strain designation name: B6.Cg-Gzmatm1Simn Gzmbtm1Ley/Apb
Genetics
Strain Types Mutant strain
Congenic
Genetic Details Targeted knockout
Mode of Inheritance Recessive
1.  Affected Gene Name: granzyme A
Chromosome 13
Affected Gene Symbol Gzma
Protein expression of altered gene: Unknown
Genetic alteration of gene: The gene was disrupted by insertion of a neomycin resistance cassette into exon 4 by homologous recombination. Western blot analysis confirmed the absence of protein product and cytochemical analysis demonstrated the absence of enzyme activity in cytotoxic T cells in homozygous mutant animals.
Location on chromosome (bp): 113884035-113891189 bp, - strand (From NCBI annotation of NCBI Build 37)

Allele Name targeted mutation 1, Markus M Simon
Allele Symbol Gzmatm1Simn
MGI Allele Accession ID MGI:2449926
Allele synonyms: GrzA-, gzm A-, gzmA-
2.  Affected Gene Name: granzyme B
Chromosome 14
Affected Gene Symbol Gzmb
Protein expression of altered gene: Nil
Genetic alteration of gene: A PGK-neomycin resistance cassette replaced a 350bp AvrII fragment starting within the 5' untranslated region (22 bp upstream from the initiation codon), extending through exon 1 and part of intron 1.
Location on chromosome (bp): 56877693-56881097 bp, - strand (From NCBI annotation of NCBI Build 37)

Allele Name targeted mutation 1, Timothy J Ley
Allele Symbol Gzmbtm1Ley
MGI Allele Accession ID MGI:1857156
Allele synonyms: Granzyme B-, GrB KO, GrzB-, gzmB-, Gzmb-/+PGK-neo
Mutant Construction Technique None
Phenotype
Phenotype Homozygous State Increased sensitivity to induced morbidity/mortality * WNV-infected double homozygotes start to die at day 10 post-infection (p.i.); wild-type mice die between 10-13 days p.i. * 70% of WNV-infected double homozygotes versus 29% of wild-type succumb to encephalitis by day 16 p.i. Immune system phenotype * double homozygotes develop a normal contact hypersensitivity (CHS) response (based on increase in ear thickness) to trinitrophenyl (TNP) relative to wild-type mice Increased susceptibility to parasitic infection: * decreased clearance and survival after infection with Trypanosoma cruzi Increased susceptibility to viral infection: * double homozygotes display increased susceptibility to West Nile flavivirus (WNV) infection relative to wild-type * double mutants show a higher level of infectious virus in brain relative to wild-type or Prf1tm1Sdz mice at the same time point
