Strain name: B6;129S4-Nos1tm1Plh/AnuApb
Nickname: Nos1 -/-
Strain designation name: B6;129S4-Nos1tm1Plh
Genetics
Strain Types Mutant strain
Genetic Details Targeted knockout
Mode of Inheritance Recessive
1.  Affected Gene Name: nitric oxide synthase 1, neuronal
Chromosome 5
Affected Gene Symbol Nos1
Protein expression of altered gene: Nil
Genetic alteration of gene: A neomycin cassette replaced exon 1. This exon encodes the initiation site and amino acids 1 - 159 of the protein. Northern blots of brain homogenates from homozygous mutant mice showed no detectable mRNA. Western blots of brain homogenates from homozygous mutant mice showed no detectable protein. However, splice variants are expressed and variable low levels of NOS activity are seen in the brain.
Location on chromosome (bp): 118231041-118408849 bp, + strand (From VEGA annotation of NCBI Build 37)

MGI:97360
Synonyms bNOS, nNOS, NO, Nos-1
Allele Name targeted mutation 1, Paul L Huang
Allele Symbol Nos1tm1Plh
MGI Allele Accession ID MGI:1857227
Allele synonyms: Kn, N2KO, nNOS KO, nNOS-, NOSdelta
Mutant Construction Technique None
Phenotype
Phenotype Homozygous State The neurons normally expressing NOS appear intact, and the mutant NOS mice are viable, fertile, and without evident histopathological abnormalities in the central nervous system. The most evident effect of disrupting the neuronal NOS gene is the development of grossly enlarged stomachs, with hypertrophy of the pyloric sphincter and the circular muscle layer. This phenotype resembles the human disorder infantile pyloric stenosis, in which gastric outlet obstruction is associated with the lack of NDP neurons in the pylorus.
