Sperm and Embryo Cryopreservation

Funding from both NHMRC and NCRIS allows us to offer subsidized mouse cryopreservation service. For strains that are in the public domain, i.e. published, or available to other researchers the full subsidy as listed below is available.

To encourage cryopreservation of unpublished strains a 2 year embargo is permitted on listing on the APB database and distribution to others. At the completion of the 2 years the embargo must be lifted or the depositor must pay for the full cryopreservation costs plus maintenance fee as outlined in the second table below.

The ongoing maintenance costs, e.g. liquid nitrogen, are also supported.

The depositor will need to pay for shipping of the mice to the APB node and the caging of the mice and breeding of mice to produce donor animals, if required.

Depositor is eligible for subsidised embryo support if:

  • All of the basic support criteria above are met and,
  • Strain cannot be frozen down as sperm due to loss of genetic integrity e.g. congenic strains.

Support does not cover

  • Production of donors, depositor is responsible or the cost of producing the required number of animals of correct sex and age,
  • Transportation of animals to cryopreservation facility
  • Strain quality control

Costs to researcher:

Sperm

Embryo

Basic subsidy*

$0

$0

Caging at cryo facility*

$0

Depends on strain, uncomplicated strain approximately $800-$1,000.

Quality control*

Subjective microscopy analysis test thaw - $0
IVF to pups - $250/strain

One validation to live pups - $0

Animal Requirements:

5 males, 10-16 weeks of age. For strains on C57BL/6 background 14 - 20 weeks of age.

The facility does not need to receive all 5 animals from the same strain at the same time.

4 males & 8 females to establish a breeding colony to provide embryo donors

Cryopreservation is conducted where?

Monash University or
Australian Phenomics Facility (ANU)

Animal Resource Centre (Perth)

Both cryopreservation services are requested through and organized by the APB Curator. The researcher does not need to do anything other than supply strain and animal information and ensure the mice arrive at the cryopreservation site on time.

Re-animation service

Where

ANU or Monash University
IVF or Intra Cytoplasmic Sperm Injection (ICSI)

(ICSI is available at Monash University only).

All centres.
If embryos were frozen down at ARC they will be re-animated at ARC.

Cost*

$350 - 3x frozen straws
$2,500 + cage costs for weaned pups

from $1925 (incl GST) (includes up to 4 thaw and transfer sessions)

For those strains that are not freely distributable depositors will be charged for the cryopreservation, on going maintenance (a yearly fee) and for shipment.

Cryopreservation* $750 (5 mice, per strain) $2650 (incl GST) (includes up to 10 freezing sessions)

Quality Control*

Subjective microscopy analysis test thaw - $60/10 mice

IVF to blastocyst - $450/strain

Test thaw and incubation to blastocyst stage - $165/strain

Yearly maintenance fee*

$100 + GST per strain $136.50 (incl GST)
